《惡靈古堡 5》類型為動作冒險，遊戲故事設定在 1 代洋房事件 10 年後，主角為原 S.T.A.R.S 隊員 克里斯（Chris），他曾在 1 代中擔任主角，以及在《惡靈古堡：聖女密碼》中現身幫助妹妹克蕾兒（Claire）。 遊戲故事敘述看似非洲乾旱荒野地區的某地，發生了大規模的衝突與紛爭，名為「BSAA」的組織於是派遣克里斯來到當地進行調查。這次克里斯所面對的敵人並不是殭屍或寄牲獸，而是外觀貌似人類且具備智能，但是行動卻血腥殘暴的大批群眾。 遊戲承襲《惡靈古堡 4》的基本系統，同樣採用第三人稱背後視點的動作冒險方式呈現，並加入目前尚未公布詳情的全新系統。不同於先前系列作慣用的黑暗場景未知恐懼感，本代將以明暗激烈轉變下所造成的短暫視力喪失，來展現另一種不同的恐懼感。
